Rebalance Illumina Libraries
This tool helps you rebalance a pooled sequencing library so under-represented samples contribute more and over-represented samples contribute less in the next pool. You paste each sample’s observed % reads, choose a desired input volume (defaults to 21 µL), and the tool calculates a rebalancing factor for each sample as (highest % reads in the cohort) ÷ (sample’s % reads), so lower-percentage samples get larger factors. Volumes are then normalized so the largest resulting volume equals your chosen desired volume, and all other volumes are scaled proportionally. Since volume of libraries is the limiting factor, we use the input desired volume to ensure there is sufficient material for the lowest-factor sample. The output table reports, for each sample, its % reads, the rebalancing factor, and the recommended input volume (to one decimal place). This tool is for investigational use only and has not been validated for clinical settings.
